Research Area:

Life cycle of the red blood cells in mammals involves a dramatic event during which specialized cells in the bone marrow that give rise to the red blood cells loose their organelles, translational machinery and genetic material. At this stage, red blood cells become “sacks” filled predominantly with a macromolecules involved in transport and delivery of oxygen from lungs to the tissues. Many cellular catalysts are involved in coordination and execution of the above-mentioned event; one of them is pyrimidine 5´-nucleotidase type 1. This enzyme is responsible for a selective cleanup of potentially toxic chemicals derived from degradation of translational machinery. When this enzyme is faulty (e.g. due to a genetic mutation or when it becomes inhibited due to lead poisoning), the subjects will suffer from a mild-to-moderate anemia, described in medical literature as hereditary pyrimidine 5´-nucleotidase deficiency. Our current research is aimed to elucidate how does this enzyme recognize its substrate(s) (pyrimidine nucleotides) and how does it discriminate against abundant substrate-like chemicals (purine nucleotides) that are produced during the degradation of translational machinery material.
